Medici王朝崛起掌權期間，正是在此安身立業。Cosimo de' Medici於1444年委託Michelozzo興建此宮，其立面由下而上漸趨平滑的粗琢石工手法，日後成為文藝復興宮殿建築的典範，並在意大利各地廣受仿效。Lorenzo the Magnificent自幼在此成長，數十年間，這裡一直是佛羅倫斯的政治神經中樞，直到Medici家族遷往Palazzo Vecchio，後又移居Palazzo Pitti。 館內最不容錯過的亮點，是位於一樓的賢士禮拜堂——這座小巧私密的禮拜堂中，Benozzo Gozzoli於1450年代末繪製了一組非凡絕倫的壁畫系列，描繪「賢士行列」的場景。畫中色彩鮮艷得令人難以置信，細節之精妙更是驚人（Medici家族成員亦以行列中人物的身份現身畫中），加上空間的私密感，讓人恍如置身於一幅立體呈現的泥金手抄本之中。每5分鐘僅限10人入內參觀，因此空間內從不會擁擠。宮內另設有Galleria degli Specchi（鏡廳），這座巴洛克式代表作以Luca Giordano繪製的天頂壁畫為亮點，此外亮還設有考古展覽路線及不定期更換的臨時展覽。 相比Uffizi或Pitti，這裡的參觀規模小得多，主題也更為集中——大約一小時左右便可細細看完。但單是Gozzoli的禮拜堂壁畫，已值得專程前來；而這座宮殿所呈現的Medici家族日常生活氣息，也是其他更宏大的博物館難以比擬的。
