Primo museo pubblico di Berlino, progettato da Karl Friedrich Schinkel e inaugurato nel 1830, l'edificio si affaccia sul Lustgarten con diciotto colonne ioniche e una rotonda a cupola modellata sul Pantheon di Roma. Ospita la collezione permanente dell'Antikensammlung dedicata a greci, etruschi e romani, con celebri capolavori come la Dea di Berlino, il Fanciullo orante e i ritratti di Cesare e Cleopatra. Una seconda sala, la «camera blu», espone oltre 1.300 monete antiche del Münzkabinett, mentre il piano superiore accoglie una delle più importanti raccolte etrusche al di fuori dell'Italia.
