I. M. Peis markanter Turm aus weißem Beton und der neunstöckige Glaspavillon ragen am Columbia Point unmittelbar am Wasser empor, auf einer windgepeitschten Landzunge am Hafenbecken von Dorchester. Im Inneren führt das Museum weitgehend chronologisch durch die Präsidentschaft von John F. Kennedy: vom Wahlkampf 1960 gegen Richard Nixon samt dem ersten Fernsehduell über die Amtseinführung und das Oval Office mit einer Nachbildung des Resolute Desk bis hin zur Kubakrise und dem Wettlauf um die erste bemannte Mondlandung der Amerikaner. Über den Ausstellungsräumen beherbergt das Archiv den Nachlass des 35. US-Präsidenten sowie die Ernest Hemingway Collection. Der Rundgang endet im Pavilion, einem rund 35 Meter hohen Glasatrium mit weitem Blick über die Boston Harbor Islands.
